ABSTRACT

Regulation and production of fibrinolytic enzymes from bacterial sources especially from Bacillus has taken leading role in the medical sciences for the treatment of cardiovascular disorders as it removes thrombus or clots adding to its significant role in curing human health issues saving millions. Significant progress has been made during the last few years on the studies on fibrinolytic enzymes. The present study was the isolation, production, purification and characterization of fibrinolytic enzyme from bacteria, isolated from soil near slaughter house locations. The bacterial strain coded as Bacillus sp.SCSR-0003 was employed for the production of fibrinolytic enzyme .The strain showed better production when using peptone as a nitrogen source and the enzyme activity was 1020U/ml with 25mg/ml protein concentration. The enzyme partially purified by ammonium sulphate precipitation followed by dialysis showed maximum activity at 80%, was 2292U/ml with 17.75mg/ml protein content. The partially purified fibrinolytic enzyme showed optimum temperature and pH was 30 ̊C and 9, the enzyme was stable at 40 ̊C and 7 respectively. The optimum substrate concentration for enzyme activity was 2%.In application study, this enzyme showed high blood clot lysis activity, which may be a good candidate in pharmaceutical industry.
