ABSTRACT

This work undertaken with the sample collection screened for fibrinolytic enzyme producers by observing the halo zone produced around the colonies on fibrin plate agar. Among the isolates Aspergillus carbonarius S-CSR-0007 produced largest clear zone was selected for our study. Enzyme production was optimized by testing substrates for their influence on enzyme activity. Experiments revealed that casein was the best substrate with the highest enzyme yield of 816 U/ml. The fibrinolytic enzyme was collected from the supernatant of Aspergillus carbonarius S-CSR-0007 with enzyme activity 510 U/ml and partially purified by ammonium sulfate precipitation followed by dialysis showed maximum activity at 80 %, with an enzyme activity of 1200 U/ml using tyrosine as standard. The partially purified fibrinolytic enzyme showed temperature and pH optimal activities at 45 °C and 7 respectively, and stabilities at 50°C and 8 respectively and the optimum substrate concentration was 4 %. In application study, this enzyme showed high blood clot lysis activity, which may be a good candidate in pharmaceutical industry.
