ABSTRACT

Alkaline protease production by soil isolated Bacillus Sp. was optimized in this study. Response surface methodology (RSM) was employed for the analysis of the simultaneous effect of substrate concentration, broth pH and incubation temperature on the production of alkaline protease produced by the isolate. A three-variable, three level Box-Behnken design (BBD) comprising 15 experimental runs was used to develop a second degree statistical model for the optimisation of the fermentation conditions. The optimal fermentation conditions that resulted in the maximum alkaline protease production were substrate concentration; 4%, broth pH; 10.5 and fermentation temperature; 35ºC. Under these conditions, the protease activity shown by the enzyme was 193U/mL. Validation of the model indicated only a small difference between predicted and observed values.
